I've been to Berlin 3 times. One thing I'll say, maybe cliched advice, is to get the open-top tour bus. Get the all-day ticket and hop on and off at whatever interests you most Brandenburg, Checkpoint Charlie etc.
Also, make sure you go to the east. Remarkable place. The architecture is traditional and the night-life is wild (from what I gathered). There was one corner cafe bar and I can remember the place was buzzing, the atmosphere spilling out onto the street on a Wednesday night in October. Food is also amazingly cheap and tasty.
I've been to plenty European cities but Berlin is the best out of all. It's just so diverse, so cosmopolitan, modern and historical all at once. Use the excellent public transport as much as possible to discover all this. And make sure you walk through the park in the city centre. Don't bother with the city centre too much after that, it's very average, except during Christmas.
